KPV (Lys-Pro-Val) is a naturally occurring tripeptide corresponding to the C-terminal fragment (residues 11-13) of alpha-melanocyte stimulating hormone (α-MSH). Research has demonstrated that this minimal C-terminal sequence retains the anti-inflammatory signaling properties of the full-length α-MSH without melanocortin receptor binding, suggesting a distinct mechanism of action.

Sequence
Lys-Pro-Val
Molecular Formula
C16H30N4O5
Molecular Weight
358.43 g/mol
CAS Number
67727-97-3
KPV has a molecular weight of 358.43 g/mol, making it one of the smallest bioactive peptide fragments studied. Despite lacking the pharmacophore required for melanocortin receptor (MC1R-MC5R) binding, KPV retains anti-inflammatory signaling activity, suggesting a receptor-independent mechanism possibly involving direct NF-κB pathway interaction.
Research Applications
• Inflammatory bowel research: Studies examining KPV effects in colitis models, including oral and nanoparticle-mediated delivery (Dalmasso et al., 2008).
• NF-κB pathway: Research on KPV's interaction with inflammatory transcription factor signaling.
• Dermatological research: Studies examining anti-inflammatory effects in skin cell models.
• Gut epithelial biology: Research on KPV's effects on intestinal epithelial barrier function and permeability.
Pathway Context
• NF-κB pathway inhibition (IκBα stabilization)
• IL-10 upregulation in monocyte/macrophage models
• TNF-α and IL-8 suppression in inflammatory models
• Melanocortin-independent anti-inflammatory signaling
• PepT1 transporter-mediated intestinal absorption
Preclinical Research
Key preclinical findings:
Dalmasso et al. (2008, Gastroenterology): KPV at 12.5 mg/kg/day (oral, nanoparticle-encapsulated) significantly reduced colonic inflammation in murine DSS colitis model. Reduced TNF-α by 65% and IL-6 by 58%. Identified PepT1-mediated intestinal uptake.
Brzoska et al. (2008): KPV at 1-100 µM inhibited NF-κBactivation in human keratinocytes, fibroblasts, and monocytes. IC50 for NF-κB inhibition: ~10 µM. Importantly, KPV did NOT bind melanocortin receptors (MC1R-MC5R), confirming receptor-independent mechanism.
Getting et al. (2006): KPV reduced IL-8 secretion by 50% in TNF-α-stimulated human intestinal epithelial cells at 10 µM concentration.
Typical in vitro concentrations: 1-100 µM. In vivo: 5-15 mg/kg (oral or IP).
All findings from animal or in vitro studies.

Frequently Asked Questions
KPV is verified at ≥99.5% purity via reverse-phase HPLC analysis. Each batch also undergoes ESI mass spectrometry for identity confirmation, LAL endotoxin testing (<0.1 EU/mg), and USP <71> sterility testing. A batch-specific Certificate of Analysis (COA) is included with every order.
Add bacteriostatic water (0.9% benzyl alcohol) slowly to the vial wall. Gently swirl to dissolve — do not shake or vortex. Reconstituted peptide should be stored at 2-8°C and used within 30 days. For single-use applications, sterile water may be substituted (no preservative, use immediately).
Lyophilized (powder): Store at -20°C, protected from light and moisture. Shelf life: 24 months. Reconstituted (solution): Store at 2-8°C, use within 30 days. Critical: Allow the vial to reach room temperature before opening to prevent moisture condensation inside the vial. Avoid repeated freeze-thaw cycles (max 3 recommended).
The CAS registry number for KPV is 67727-97-3. Molecular weight: 358.43 g/mol. Molecular formula: C16H30N4O5. Full chemical characterization data including PubChem identifiers, SMILES notation, and InChIKey are available on the Chemical Properties tab above.
Yes. Every batch of KPV is independently tested by Janssen Analytical Laboratories (ISO 17025-accredited). Testing includes HPLC purity analysis, mass spectrometry identity confirmation, LAL kinetic turbidimetric endotoxin assay, and USP <71> sterility testing. Full results are provided in the batch-specific COA. Additional documentation is available on the COA/HPLC/MS and 3rd Party Testing tabs above.
Orders placed before 2:00 PM EST ship same day via cold-chain logistics with gel ice packs to maintain product integrity during transit. Most domestic orders arrive within 2-4 business days. Full tracking is provided. All vials are sealed under inert atmosphere and packaged to prevent damage.
Yes. Tiered pricing is applied automatically at checkout: 2+ units at 10% off, 3+ units at 15% off, and 5+ units at 20% off. For institutional or bulk research inquiries (10+ vials), contact us at contact@aureumpeptides.com for custom pricing.
Compatibility depends on your specific experimental design. KPV is provided as a single-compound lyophilized preparation for maximum flexibility. When using multiple compounds, reconstitute each separately and add to experimental systems independently to avoid potential interactions during reconstitution.
