FOXO4-DRI is a D-retro-inverso peptide designed to disrupt the interaction between FOXO4 transcription factor and p53 in senescent cells. Developed by Peter de Keizer at Erasmus University Medical Center, FOXO4-DRI was shown to selectively induce apoptosis in senescent cells in preclinical studies, making it one of the first reported peptide-based senolytic compounds.

FOXO4-DRI is a 46-amino acid D-retro-inverso (DRI) peptide with an approximate molecular weight of 5647 g/mol. The DRI modification means all amino acids are D-enantiomers and the sequence is reversed, which preserves side-chain topology while conferring exceptional protease resistance.
The peptide is designed to competitively disrupt the FOXO4-p53 protein-protein interaction in the nucleus of senescent cells.
