CJC-1295 without DAC (Drug Affinity Complex), also known as Modified GRF 1-29 or Mod GRF, is a synthetic 29-amino acid peptide analog of growth hormone-releasing hormone (GHRH). It was developed with four amino acid substitutions to resist enzymatic degradation by dipeptidyl peptidase IV (DPP-IV), extending its biological half-life compared to native GHRH(1-29).
CJC-1295 No DAC is a tetrasubstituted analog of human GHRH(1-29) with molecular weight 3367.97 g/mol. The four amino acid substitutions at positions 2 (D-Ala), 8 (Gln), 15 (Ala), and 27 (Leu) confer resistance to DPP-IV proteolysis.
Unlike the DAC-conjugated variant, this form does not bind albumin and has a shorter circulating half-life, making it useful for pulsatile GHRH receptor activation studies.
Research Applications
• GHRH receptor biology: Studies have examined CJC-1295's interaction with the GHRH receptor (GHRHR) and downstream somatotroph signaling.
• GH secretion research: Investigations into pulsatile GH release patterns and the synergistic effects with ghrelin-mimetic peptides.
• Metabolic research: Studies examining GH axis modulation and associated effects on lipolysis and nitrogen balance in animal models.
• Neuroendocrine research: Investigations into hypothalamic-pituitary axis feedback mechanisms.
Pathway Context
• GHRHR activation → cAMP/PKA signaling cascade
• Somatotroph cell GH synthesis and pulsatile release
• IGF-1 axis downstream signaling (hepatic)
• Synergistic interaction with ghrelin receptor (GHS-R1a) pathway when co-administered with GHRP analogs
Preclinical Research
Teichman et al. (2006) characterized dose-dependent GH release dynamics following subcutaneous CJC-1295 administration in Phase I clinical pharmacology research.
Alba et al. (2006) showed sustained elevations in GH and IGF-1 levels in healthy volunteers with CJC-1295 (DAC-conjugated form).
Note: These studies used the DAC-conjugated form. Non-DAC research is primarily preclinical.

Frequently Asked Questions
CJC-1295 No DAC is verified at ≥99.1% purity via reverse-phase HPLC analysis. Each batch also undergoes ESI mass spectrometry for identity confirmation, LAL endotoxin testing (<0.1 EU/mg), and USP <71> sterility testing. A batch-specific Certificate of Analysis (COA) is included with every order.
Add bacteriostatic water (0.9% benzyl alcohol) slowly to the vial wall. Gently swirl to dissolve — do not shake or vortex. Reconstituted peptide should be stored at 2-8°C and used within 30 days. For single-use applications, sterile water may be substituted (no preservative, use immediately).
Lyophilized (powder): Store at -20°C, protected from light and moisture. Shelf life: 24 months. Reconstituted (solution): Store at 2-8°C, use within 30 days. Critical: Allow the vial to reach room temperature before opening to prevent moisture condensation inside the vial. Avoid repeated freeze-thaw cycles (max 3 recommended).
