Telomerase Activation in Cell Culture

Primary human fibroblasts or lymphocytes cultured to near-replicative senescence (late passages) — Epitalon exposure at nM–µM concentrations followed by TRAP assay (telomeric repeat amplification protocol) for telomerase activity quantification.

Telomere Length Measurement

Terminal restriction fragment (TRF) Southern blot, or qPCR-based telomere length measurement, baseline to follow-up extended Epitalon exposure in culture. Published reports describe modest telomere length preservation.

Hayflick Limit / Replicative Senescence

Extended culture of diploid fibroblasts past ~40 population doublings with periodic Epitalon treatment — assessment of continued proliferation capacity, senescence markers (SA-β-gal), and DNA damage markers (γH2AX foci).

Aged Rodent Lifespan Studies

Aged mice (18+ months) with periodic Epitalon administration followed to natural death for lifespan analysis. Secondary endpoints: age-related biomarkers (GDF-15, IGF-1, inflammatory cytokines), tissue-level oxidative stress markers.

Research Considerations

Epitalon’s reported effects are generally modest and require sensitive measurement techniques. Studies with small sample sizes or coarse endpoints frequently fail to detect effects. Statistical power considerations are critical.
